RESUMEN

Plantar pressure measurement systems are routinely used in sports and health applications to assess locomotion. The purpose of this review is to describe and critically discuss: (a) applications of the pressure measurement systems in sport and healthcare, (b) testing protocols and considerations for clinical gait analysis, (c) clinical recommendations for interpreting plantar pressure data, (d) calibration procedures and their accuracy, and (e) the future of pressure sensor data analysis. Rigid pressure platforms are typically used to measure plantar pressures for the assessment of foot function during standing and walking, particularly when barefoot, and are the most accurate for measuring plantar pressures. For reliable data, two step protocol prior to contacting the pressure plate is recommended. In-shoe systems are most suitable for measuring plantar pressures in the field during daily living or dynamic sporting movements as they are often wireless and can measure multiple steps. They are the most suitable equipment to assess the effects of footwear and orthotics on plantar pressures. However, they typically have lower spatial resolution and sampling frequency than platform systems. Users of pressure measurement systems need to consider the suitability of the calibration procedures for their chosen application when selecting and using a pressure measurement system. For some applications, a bespoke calibration procedure is required to improve validity and reliability of the pressure measurement system. The testing machines that are commonly used for dynamic calibration of pressure measurement systems frequently have loading rates of less than even those found in walking, so the development of testing protocols that truly measure the loading rates found in many sporting movements are required. There is clear potential for AI techniques to assist in the analysis and interpretation of plantar pressure data to enable the more complete use of pressure system data in clinical diagnoses and monitoring.

PURPOSE: To determine the effect of taurine supplementation on sweating and core temperature responses, including the transition from compensable to uncompensable heat stress, during prolonged low-intensity exercise of a fixed-heat production (~ 200W/m2) in hot conditions (37.5 °C), at both fixed and incremental vapour-pressure. METHODS: Fifteen females (n = 3) and males (n = 12; 27 ± 5 years, 78 ± 9 kg, V ˙ O2max 50.3 ± 7.8 mL/kg/min), completed a treadmill walking protocol (~ 200W/m2 heat production [Hprod]) in the heat (37.5 ± 0.1 °C) at fixed-(16-mmHg) and ramped-humidity (∆1.5-mmHg/5-min) following 1 week of oral taurine supplementation (50 mg/kg/bm) or placebo, in a double-blind, randomised, cross-over design. Participants were assessed for whole-body sweat loss (WBSL), local sweat rate (LSR), sweat gland activation (SGA), core temperature (Tcore), breakpoint of compensability (Pcrit) and calorimetric heat transfer components. Plasma volume and plasma taurine concentrations were established through pre- and post-trial blood samples. RESULTS: Taurine supplementation increased WBSL by 26.6% and 5.1% (p = 0.035), LSR by 15.5% and 7.8% (p = 0.013), SGA (1 × 1 cm) by 32.2% and 29.9% (p < 0.001) and SGA (3 × 3 cm) by 22.1% and 17.1% (p = 0.015) during the fixed- and ramped-humidity exercise periods, respectively. Evaporative heat loss was enhanced by 27% (p = 0.010), heat-storage reduced by 72% (p = 0.024) and Pcrit was greater in taurine vs placebo (25.0-mmHg vs 21.7-mmHg; p = 0.002). CONCLUSION: Taurine supplementation increased sweating responses during fixed Hprod in hot conditions, prior to substantial heat strain and before the breakpoint of compensability, demonstrating improved thermoregulatory capacity. The enhanced evaporative cooling and reduced heat-storage delayed the subsequent upward inflection in Tcore-represented by a greater Pcrit-and offers a potential dietary supplementation strategy to support thermoregulation.

PURPOSE: Mental rehearsal is commonly employed, with positive visualization proposed to enhance complex skill performance. Additionally, video stimulus has been associated with enhanced kinesthetic sensations and rapid hormone fluctuations that may contribute to enhancing mental rehearsal and the conscious and unconscious emotional state for skill execution. Here, we assessed the impact of a 15-minute mental rehearsal intervention on rugby-specific tasks and the associated hormone profile. METHODS: Professional rugby players (N = 10) volunteered for a randomized crossover study. They completed three 15-minute preparatory phases (positive or negative video-guided mental rehearsal or self-directed mental rehearsal alone) prior to an exercise stressor and rugby-specific passing task. Salivary testosterone and cortisol were monitored to assess stress responses. RESULTS: Performance during the rugby passing task was improved following the positive video condition (91% [7.4%]) compared to the negative video (79% [6.0%]; ES: 1.22 ± 0.75) and self-visualization (86% [5.8%]; ES: 0.58 ± 0.75), with a significant correlation observed between passing performance and salivary testosterone (r = .47 ± .34, P = .0087). Positive video imagery prior to an exercise stressor also significantly enhanced physiological stress resilience (r = .39 ± .36, P = .0352). CONCLUSIONS: This pilot study demonstrates that mental rehearsal was enhanced by appropriate, context-specific video presentation. We propose that the interaction between sex steroids, the adrenal axis, and subsequent conscious and unconscious behaviors may be relevant to competitive rugby. Specifically, we suggest that relatively elevated free testosterone imparts a degree of stress resilience, which may lead to enhanced expression of competitive behaviors and provide an enhanced state for rugby skill execution.

RESUMEN

Pressure measurement systems have numerous applications in healthcare and sport. The purpose of this review is to: (a) describe the brief history of the development of pressure sensors for clinical and sport applications, (b) discuss the design requirements for pressure measurement systems for different applications, (c) critique the suitability, reliability, and validity of commercial pressure measurement systems, and (d) suggest future directions for the development of pressure measurements systems in this area. Commercial pressure measurement systems generally use capacitive or resistive sensors, and typically capacitive sensors have been reported to be more valid and reliable than resistive sensors for prolonged use. It is important to acknowledge, however, that the selection of sensors is contingent upon the specific application requirements. Recent improvements in sensor and wireless technology and computational power have resulted in systems that have higher sensor density and sampling frequency with improved usability - thinner, lighter platforms, some of which are wireless, and reduced the obtrusiveness of in-shoe systems due to wireless data transmission and smaller data-logger and control units. Future developments of pressure sensors should focus on the design of systems that can measure or accurately predict shear stresses in conjunction with pressure, as it is thought the combination of both contributes to the development of pressure ulcers and diabetic plantar ulcers. The focus for the development of in-shoe pressure measurement systems is to minimise any potential interference to the patient or athlete, and to reduce power consumption of the wireless systems to improve the battery life, so these systems can be used to monitor daily activity. A potential solution to reduce the obtrusiveness of in-shoe systems include thin flexible pressure sensors which can be incorporated into socks. Although some experimental systems are available further work is needed to improve their validity and reliability.

PURPOSE: The efficacy of isolated and relative performance indicators (PIs) has been compared in rugby union; the latter more effective at discerning match outcomes. However, this methodology has not been applied in women's rugby. The aim of this study was to identify PIs that maximize prediction accuracy of match outcome, from isolated and relative data sets, in women's rugby union. METHODS: Twenty-six PIs were selected from 110 women's international rugby matches between 2017 and 2022 to form an isolated data set, with relative data sets determined by subtracting corresponding opposition PIs. Random forest classification was completed on both data sets, and feature selection and importance were used to simplify models and interpret key PIs. Models were used in prediction on the 2021 World Cup to evaluate performance on unseen data. RESULTS: The isolated full model correctly classified 75% of outcomes (CI, 65%-82%), whereas the relative full model correctly classified 78% (CI, 69%-86%). Reduced respective models correctly classified 74% (CI, 65%-82%) and 76% (CI, 67%-84%). Reduced models correctly predicted 100% and 96% of outcomes for isolated and relative test data sets, respectively. No significant difference in accuracy was found between data sets. In the relative reduced model, meters made, clean breaks, missed tackles, lineouts lost, carries, and kicks from hand were significant. CONCLUSIONS: Increased relative meters made, clean breaks, carries, and kicks from hand and decreased relative missed tackles and lineouts lost were associated with success. This information can be utilized to inform physical and tactical preparation and direct physiological studies in women's rugby.

RESUMEN

PURPOSE: As the start of the 10th Rugby Union World Cup approaches, performance staff will be working on the final elements of their teams' preparation. Much of this planning and preparation will be underpinned by the latest performance science research. In this invited commentary, we discuss contemporary performance science research in rugby union centered around 4 key performance domains. First, we outline a systematic approach to developing an overall understanding of the game demands and how performance staff can enhance the players' preparedness for competition. We then move on to outline our understanding of the training science domain, followed by a brief overview of effective recovery strategies at major tournaments. Finally, we outline research in the area of competition-day strategies and how they can positively impact players' readiness to compete. CONCLUSIONS: Evaluating a team's preparation for the Rugby Union World Cup can be achieved by mapping their performance plan based on the 4 domains outlined above.

PURPOSE: This study aimed to determine the effects of a field-based priming session on perceptual, physiological, and performance responses in female rugby sevens athletes. METHODS: Thirteen highly trained female rugby sevens players (age: 20.7 [2.0] y; height: 169.3 [4.8] cm; weight: 68.8 [7.9] kg) completed either a 20-minute field-based priming session or a control condition. Perceptual, physiological, and performance variables were collected at baseline (PRE) and 5 (POST5), 30 (POST30), and 120 minutes (POST120) postintervention. Data were analyzed using Bayesian mixed effects models. RESULTS: The priming protocol had a larger increase in mental readiness (maximum a posteriori [MAP] = 20, 95% high-density intervals [HDI] = -4 to 42, probability of direction [PD]% = 95, % in region of practical equivalence [ROPE] = 9.7), physical readiness (MAP = 20.1, 95% HDI = -4.6 to 42.1, PD% = 93, % in ROPE = 10.6), and testosterone (MAP = 14.9, 95% HDI = 0.5 to 27.7, PD% = 98, % in ROPE = 5.6) than the control POST30. Cognitive performance decreased POST120 in the priming condition for congruent (MAP = 0.02, 95% HDI = -0.06 to 0.00, PD% = 95, % in ROPE = 6.4) and incongruent tasks (MAP = 0.00, 95% HDI = -0.07 to 0.00, PD% = 98, % in ROPE = 3.2) when compared with the control. CONCLUSIONS: Perceptual and physiological markers improved POST30 in the priming condition. Findings indicate that perceptual and physiological responses to priming were not coupled with performance improvements. Priming was not accompanied by perceptual, physiological, or performance improvements at POST120.

BACKGROUND: The effects of Anabolic Androgenic Steroids (AAS) are largely illustrated through Androgen Receptor induced gene transcription, yet RNA-Seq has yet to be conducted on human whole blood and skeletal muscle. Investigating the transcriptional signature of AAS in blood may aid AAS detection and in muscle further understanding of AAS induced hypertrophy. METHODS: Males aged 20-42 were recruited and sampled once: sedentary controls (C), resistance trained lifters (RT) and resistance trained current AAS users (RT-AS) who ceased exposure ≤ 2 or ≥ 10 weeks prior to sampling. RT-AS were sampled twice as Returning Participants (RP) if AAS usage ceased for ≥ 18 weeks. RNA was extracted from whole blood and trapezius muscle samples. RNA libraries were sequenced twice, for validation purposes, on the DNBSEQ-G400RS with either standard or CoolMPS PE100 reagents following MGI protocols. Genes were considered differentially expressed with FDR < 0.05 and a 1.2- fold change. RESULTS: Cross-comparison of both standard reagent whole blood (N = 55: C = 7, RT = 20, RT-AS ≤ 2 = 14, RT-AS ≥ 10 = 10, RP = 4; N = 46: C = 6, RT = 17, RT-AS ≤ 2 = 12, RT-AS ≥ 10 = 8, RP = 3) sequencing datasets, showed that no genes or gene sets/pathways were differentially expressed between time points for RP or between group comparisons of RT-AS ≤ 2 vs. C, RT, or RT-AS ≥ 10. Cross-comparison of both muscle (N = 51, C = 5, RT = 17, RT-AS ≤ 2 = 15, RT-AS ≥ 10 = 11, RP = 3) sequencing (one standard & one CoolMPS reagent) datasets, showed one gene, CHRDL1, which has atrophying potential, was upregulated in RP visit two. In both muscle sequencing datasets, nine differentially expressed genes, overlapped with RT-AS ≤ 2 vs. RT and RT-AS ≤ 2 vs. C, but were not differentially expressed with RT vs. C, possibly suggesting they are from acute doping alone. No genes seemed to be differentially expressed in muscle after the long-term cessation of AAS, whereas a previous study found long term proteomic changes. CONCLUSION: A whole blood transcriptional signature of AAS doping was not identified. However, RNA-Seq of muscle has identified numerous differentially expressed genes with known impacts on hypertrophic processes that may further our understanding on AAS induced hypertrophy. Differences in training regimens in participant groupings may have influenced results. Future studies should focus on longitudinal sampling pre, during and post-AAS exposure to better control for confounding variables.

Day-to-day coordination of the stress (i.e., hypothalamic-pituitary-adrenal [HPA]) and reproductive (i.e., hypothalamic-pituitary-gonadal [HPG]) axes is central to allostatic regulation, reproductive success, and survival. Reports of positive, within-person testosterone and cortisol relationships (or coupling) suggest cross-talk of a facilitative nature, but longitudinal evidence is scarce and has methodological and analytical limitations. To address this, we used a continuous-time (CT) model to investigate day-to-day, within-person coupling of testosterone and cortisol in two male cohorts. Salivary testosterone and cortisol fluctuations were monitored in 35 athletic men across two international tournaments (M = 19.3 tests) and in 41 healthy men during normal daily living (M = 27.9 tests). Bayesian CT analysis revealed a diminishing effect of each hormone on itself as time-interval length or lag increased. In both groups, cortisol had a negative lagged effect on testosterone that persisted for around three days. The cortisol effect on testosterone peaked after 0.71 and 0.51 days in athletic (standardized estimate = -0.13) and healthy men (standardized estimate = -0.11), respectively. Further estimates of non-lagged, contemporaneous correlations revealed positive testosterone and cortisol relationships (athlete r = 0.04, healthy r = 0.46). In summary, complex within-person HPA and HPG interplay emerged in two independent male cohorts. Specifically, a rising cortisol concentration was linked to a fall in testosterone concentration at later time points, but concurrently these hormones tended to rise and fall together. Our results suggest that inhibitory and facilitatory hormonal actions coexist on varying timescales, thereby expanding knowledge of HPG and HPA cross-talk in everyday life.

Part 1 of this genetic association series highlighted several genetic variants independently associated with elite status in rugby. However, it is highly likely that the genetic influence on elite status is polygenic due to the interaction of multiple genes. Therefore, the aim of the present study was to investigate whether polygenic profiles of elite rugby athletes differed from non-athletes utilising 13 genetic polymorphisms previously associated with tendon/ligament injury. Total genotype score (TGS) was calculated and multifactor dimensionality reduction (MDR) was used to calculate SNP-SNP epistasis interactions. Based on our elite rugby data from Part 1, mean TGS was significantly higher in elite rugby athletes (52.1 ± 10.7) than non-athletes (48.7 ± 10.8). There were more elite rugby athletes (54%) within the upper TGS quartile, and fewer (46%) within the lower quartile, compared to non-athletes (31% and 69%, respectively; P = 5·10-5), and the TGS was able to distinguish between elite rugby athletes and non-athletes (area under the curve = 0.59; 95% confidence interval 0.55-0.63; P = 9·10-7). Furthermore, MDR identified a three-SNP model of COL5A1 rs12722, COL5A1 rs3196378 and MIR608 rs4919510 that was best able to predict elite athlete status, with a greater frequency of the CC-CC-CC genotype combination in elite rugby athletes (9.8%) than non-athletes (5.3%). We propose that elite rugby athletes possess "preferable" musculoskeletal soft-tissue injury-associated polygenic profiles that have helped them achieve success in the high injury risk environment of rugby. These data may, in future, have implications for the individual management of musculoskeletal soft-tissue injury.HighlightsElite rugby athletes have preferable polygenic profiles to non-athletes in terms of genetic variants previously associated with musculoskeletal soft-tissue injury.The total genotype score was able to distinguish between elite rugby athletes and non-athletes.COL5A1 rs12722, COL5A1 rs3196378 and MIR608 rs4919510 produced the best model for predicting elite athlete status.We propose that elite rugby athletes may have an inherited advantage to achieving elite status due to an increased resistance to soft-tissue injury.

OBJECTIVES: The aims of this study were to: i) identify performance indicators associated with match outcomes in the United Rugby Championship; ii) compare the efficacy of isolated and relative datasets to predict match outcome; and iii) investigate whether reduced statistical models can reproduce predictive accuracy. DESIGN: Retrospective analysis of key performance indicators in the United Rugby Championship. METHODS: Twenty-seven performance indicators were selected from 96 matches (2020-21 United Rugby Championship). Random forest classification was completed on isolated and relative datasets, using a binary match outcome (win/lose). Maximum relevance and minimum redundancy performance indicator selection was utilised to reduce models. In addition, models were tested on 53 matches from the 2021-22 season to ascertain prediction accuracy. RESULTS: Within the 2020-21 datasets, the full models correctly classified 83% of match performances for the relative dataset and 64% for isolated data, the equivalent reduced models classified 85% and 66% respectively. The reduced relative model successfully predicted 90% of match performances in the 21-22 season, highlighting that five performance indicators were significant: kicks from hand, metres made, clean breaks, turnovers conceded and scrum penalties. CONCLUSIONS: Relative performance indicators were more effective in predicting match outcomes than isolated data. Reducing features used in random forest classification did not degrade prediction accuracy, whilst also simplifying interpretation for practitioners. Increased kicks from hand, metres made, and clean breaks compared to the opposition, as well as fewer scrum penalties and turnovers conceded were all indicators of winning match outcomes within the United Rugby Championship.

Alternate leg bounding acutely improves change-of-direction performance in women's team sports players irrespective of ground type. J Strength Cond Res 37(6): 1199-1203, 2023-This study aimed to assess whether body mass only alternate leg bounding performed post-warm-up on grass or a hard surface acutely improves preplanned change-of-direction performance in women's team sports players relative to a control condition and, if so, profile the time course of such changes. On 3 occasions, 14 amateur women's team sports players performed 20 m preplanned change-of-direction ("Pro-Agility") tests at 4, 8, and 12 minutes after interventions. Interventions were implemented immediately after a standardized warm-up and consisted of 3 sets of 10 repetitions of alternate leg bounding (5 ground contacts per limb) on a hard indoor surface (HARD) or natural grass (GRASS), or a control condition involving approximately 75 seconds of continuous walking with no bounding (CON). Performance was similar between conditions at 4-minutes postintervention. Performance at 8 minutes was greater in HARD (2.9%, p = 0.015) and GRASS (3.8%, p = 0.029) relative to CON, whereas GRASS also exceeded CON at 12 minute post-bounding (5.2%, p = 0.004). All effects were large. No differences existed between HARD and GRASS at any time. Irrespective of the ground surface, alternate leg bounding performed with body mass only can acutely improve indices of change-of-direction performance in women's team sports players when an appropriate post-stimulus recovery period is provided. Bounding on grass or a hard surface represents a feasible match-day practice that enhances subsequent change-of-direction performance and could therefore be used as part of practically applicable pre-match, half-time, and pitch-side (re)warm-up activities.

This study assessed the influence of environmental factors, air travel, and epoch estimation method on locomotor demands of international men's rugby sevens match-play. Eighteen men's rugby sevens players wore 10 Hz Global Positioning Systems (STATsport) during 52 international matches over nine global tournaments (418 observations). Whole-match average speed was recorded, whilst average speed and relative high-speed distance (>5.0 m·s-1) were quantified using FIXED and ROLL methods over 60-420 s epochs (60 s increments) to establish worst-case scenario demands. Linear mixed models compared FIXED versus ROLL estimation methods and assessed whether temperature, humidity, travel duration, number of time-zones crossed, and travel direction were associated with locomotor responses. Temperature and humidity were positively associated with overall and worst-case scenario average speed (effect estimates; b: 0.18-0.54), whilst worst-case scenario high-speed distance at 300 s was also related to temperature (b: 0.19). Easterly air travel compromised overall and 180 and 300 s worst-case scenario average speed (b: -8.31 to-7.39), alongside high-speed distance over 300 s (b: -4.54). For worst-case scenario average speed and high-speed distance, FIXED underestimated ROLL at all epoch lengths (∼9.9-18.4%, p≤0.001). This study indicated that international rugby sevens match-play locomotor responses were greater as air temperature increased but reduced following eastward air travel. Underestimation of demands in FIXED vs ROLL over 60-420 s epochs was confirmed. Such climatic and travel influences warrant the adoption of strategies targeted at maximising performance and safety according to the tournament conditions. Knowing the most demanding periods of match-play facilitates training specificity.HighlightsSelected locomotor responses were reduced following eastward air travel, potentially suggesting interventions to mitigate these effects are warranted.Match-play running responses were greater as air temperature increased. Strategies targeted at optimising body temperature in both warm and cool conditions warrant consideration to promote performance and maintain player safety.Fixed epochs underestimated worst-case scenario average speed (9.9-11.7%) and high-speed distance (11.4-18.4%). Rolling averages may thus be more appropriate for detecting the most intense periods, while duration-specific data provide training targets.

There is growing evidence of genetic contributions to tendon and ligament pathologies. Given the high incidence and severity of tendon and ligament injuries in elite rugby, we studied whether 13 gene polymorphisms previously associated with tendon/ligament injury were associated with elite athlete status. Participants from the RugbyGene project were 663 elite Caucasian male rugby athletes (RA) (mean (standard deviation) height 1.85 (0.07) m, mass 101 (12) kg, age 29 (7) yr), including 558 rugby union athletes (RU) and 105 rugby league athletes. Non-athletes (NA) were 909 Caucasian men and women (56% female; height 1.70 (0.10) m, mass 72 (13) kg, age 41 (23) yr). Genotypes were determined using TaqMan probes and groups compared using Χ2 and odds ratio (OR). COLGALT1 rs8090 AA genotype was more frequent in RA (27%) than NA (23%; P = 0.006). COL3A1 rs1800255 A allele was more frequent in RA (26%) than NA (23%) due to a greater frequency of GA genotype (39% vs 33%). For MIR608 rs4919510, RA had 1.7 times the odds of carrying the CC genotype compared to NA. MMP3 rs591058 TT genotype was less common in RA (25.1%) than NA (31.2%; P < 0.04). For NID1 rs4660148, RA had 1.6 times the odds of carrying the TT genotype compared to NA. It appears that elite rugby athletes have an inherited advantage that contributes to their elite status, possibly via resistance to soft tissue injury. These data may, in future, assist personalised management of injury risk amongst athletes.Highlights The elite rugby athletes we studied had differing genetic characteristics to non-athletes regarding genetic variants previously associated with soft-tissue injury risk.COLGALT1 rs8090, COL3A1 rs1800255, MIR608 rs4919510, MMP3 rs591058 and NID1 rs4660148 were all associated with elite status in rugby.We propose that elite rugby athletes might possess an inherited resistance to soft tissue injury, which has enabled them to achieve elite status despite exposure to the high-risk environment of elite rugby.

OBJECTIVE: It remains unknown whether myonuclei remain elevated post anabolic-androgenic steroid (AAS) usage in humans. Limited data exist on AAS-induced changes in gene expression. DESIGN: Cross-sectional/longitudinal. SETTING: University. PARTICIPANTS: Fifty-six men aged 20 to 42 years. INDEPENDENT VARIABLES: Non-resistance-trained (C) or resistance-trained (RT), RT currently using AAS (RT-AS), of which if AAS usage ceased for ≥18 weeks resampled as Returning Participants (RP) or RT previously using AAS (PREV). MAIN OUTCOME MEASURES: Myonuclei per fiber and cross-sectional area (CSA) of trapezius muscle fibers. RESULTS: There were no significant differences between C (n = 5), RT (n = 15), RT-AS (n = 17), and PREV (n = 6) for myonuclei per fiber. Three of 5 returning participants (RP1-3) were biopsied twice. Before visit 1, RP1 ceased AAS usage 34 weeks before, RP2 and RP3 ceased AAS usage ≤2 weeks before, and all had 28 weeks between visits. Fiber CSA decreased for RP1 and RP2 between visits (7566 vs 6629 µm 2 ; 7854 vs 5677 µm 2 ) while myonuclei per fiber remained similar (3.5 vs 3.4; 2.5 vs 2.6). Respectively, these values increased for RP3 between visits (7167 vs 7889 µm 2 ; 2.6 vs 3.3). CONCLUSIONS: This cohort of past AAS users did not have elevated myonuclei per fiber values, unlike previous research, but reported AAS usage was much lower. Training and AAS usage history also varied widely among participants. Comparable myonuclei per fiber numbers despite decrements in fiber CSA postexposure adheres with the muscle memory mechanism, but there is variation in usage relative to sampling date and low numbers of returning participants.

OBJECTIVE: Concussions are common match injuries in elite rugby, and reports exist of reduced cognitive function and long-term health consequences that can interrupt or end a playing career and produce continued ill health. The aim of this study was to investigate the association between elite rugby status and 8 concussion-associated risk polymorphisms. We hypothesized that concussion-associated risk genotypes and alleles would be underrepresented in elite rugby athletes compared with nonathletes. DESIGN: A case-control genetic association study. SETTING: Institutional (university). PARTICIPANTS: Elite White male rugby athletes [n = 668, mean (SD) height 1.85 (0.07) m, mass 102 (12) kg, and age 29 (7) years] and 1015 nonathlete White men and women (48% men). INTERVENTIONS: Genotype was the independent variable, obtained by PCR of genomic DNA using TaqMan probes. MAIN OUTCOME MEASURE: Elite athlete status with groups compared using χ 2 and odds ratio (OR). RESULTS: The COMT rs4680 Met/Met (AA) genotype, Met allele possession, and Met allele frequency were lower in rugby athletes (24.8%, 74.6%, and 49.7%, respectively) than nonathletes (30.2%, 77.6%, and 54.0%; P < 0.05). The Val/Val (GG) genotype was more common in elite rugby athletes than nonathletes (OR 1.39, 95% confidence interval 1.04-1.86). No other polymorphism was associated with elite athlete status. CONCLUSIONS: Elite rugby athlete status is associated with COMT rs4680 genotype that, acting pleiotropically, could affect stress resilience and behavioral traits during competition, concussion risk, and/or recovery from concussion. Consequently, assessing COMT rs4680 genotype might aid future individualized management of concussion risk among athletes.

OBJECTIVE: Inter-day reliability of sweat measurements, including the absorbent patch and modified iodine-paper techniques, at rest and exercise were evaluated. We further evaluated the effect of iodine paper size and the method of establishing sweat gland activation (sweat gland counting or surface area covered) on reliability. Furthermore, the relationships between all measurement techniques and metabolic heat production [Hprod] and evaporative requirement for heat balance [Ereq] were determined. METHOD: Twelve participants were assessed for whole-body sweat loss (WBSL), local sweat rate (LSR; absorbent patch) and sweat gland activation (SGA; iodine-paper) during rest and sub-maximal cycling at ~200, ~250 and ~300 W/m2 Hprod in the heat. Variations in iodine paper (1 x 1 cm-9 x 9 cm) were used to quantify SGA by counting sweat glands or surface area covered. The 'optimal' area of SGA was also determined based on the highest density of recruited glands. RESULTS: All measures of the sweating response were positively related with Hprod and Ereq (r = 0.53-0.84), with the 9 x 9 cm and 6 x 6 cm iodine paper sizes being the strongest (r = 0.66-0.84) for SGA. Superior inter-day reliability was found for all measures during exercise (CV% = 6-33.2) compared to rest (CV% = 33.5-77.9). The iodine-paper technique was most reliable at 9 x 9 cm (CV% = 15.9) or when the 1 x 1 cm (CV% = 17.6) and 3 x 3 cm (CV% = 15.5) optimal SGA was determined, particularly when measuring the sweat gland number. SIGNIFICANCE: WBSL, LSR and SGA measurement techniques are sufficiently reliable to detect changes in thermal sweating typically reported. We recommend 9 x 9 cm paper sizes or 1 x 1 cm-3 x 3 cm optimal areas, using either gland counting or surface area to determine SGA.

Due to the high-velocity collision-based nature of elite rugby league and union, the risk of sustaining a concussion is high. Occurrence of and outcomes following a concussion are probably affected by the interaction of multiple genes in a polygenic manner. This study investigated whether suspected concussion-associated polygenic profiles of elite rugby athletes differed from non-athletes and between rugby union forwards and backs. We hypothesised that a total genotype score (TGS) using eight concussion-associated polymorphisms would be higher in elite rugby athletes than non-athletes, indicating selection for protection against incurring or suffering prolonged effects of, concussion in the relatively high-risk environment of competitive rugby. In addition, multifactor dimensionality reduction was used to identify genetic interactions. Contrary to our hypothesis, TGS did not differ between elite rugby athletes and non-athletes (p ≥ 0.065), nor between rugby union forwards and backs (p = 0.668). Accordingly, the TGS could not discriminate between elite rugby athletes and non-athletes (AUC ~0.5), suggesting that, for the eight polymorphisms investigated, elite rugby athletes do not have a more 'preferable' concussion-associated polygenic profile than non-athletes. However, the COMT (rs4680) and MAPT (rs10445337) GC allele combination was more common in rugby athletes (31.7%; p < 0.001) and rugby union athletes (31.8%; p < 0.001) than non-athletes (24.5%). Our results thus suggest a genetic interaction between COMT (rs4680) and MAPT (rs10445337) assists rugby athletes in achieving elite status. These findings need exploration vis-à-vis sport-related concussion injury data and could have implications for the management of inter-individual differences in concussion risk.

The 24 h responses to professional female netball-specific training were examined. British Superleague players (n = 14) undertook a 90-min on-court training session incorporating key movement, technical, and scenario-specific match-play drills. Perceptual (mood, fatigue, soreness), neuromuscular (countermovement jump peak power output [PPO], PPO relative to mass [PPOrel], jump height [JH]), endocrine (salivary cortisol [C], testosterone [T] concentrations) and biochemical (creatine kinase concentrations [CK]) markers were assessed at baseline (immediately before; Pre), and immediately, two and 24 hours after (+0h, +2h, +24h) training. Session (sRPE) and differential (dRPE) ratings of perceived exertion were recorded at +0h. Identification of clear between time-point differences were based on the 95% confidence interval (CI) for mean differences relative to baseline values not overlapping. At +0h, C (raw unit mean difference from baseline; 95% CI: 0.16; 0.06 to 0.25 µg·dl-1), T (32; 20 to 45 pg⋅ml-1), CK (39; 28 to 50 u·L-1), PPOrel (2.4; 0.9 to 3.9 W·kg-1) and PPO (169; 52 to 286 W) increased. At +2h, fatigue (15; 7 to 24 AU), CK (49; 38 to 60 u·L-1), and soreness (14; 3 to 25 AU) increased, while T (-24; -37 to -11 pg⋅ml-1) and mood (-20; -27 to -12 AU) reduced. At +24h, CK increased (25; 13 to 36 u·L-1) whereas PPOrel (-1.6; -3.2 to -0.1 W·kg-1) and JH (-0.02; -0.03 to -0.08 m) reduced. Responses were variable specific, and recovery of all variables did not occur within 24h. The residual effects of the prior stimulus should be accounted for in the planning of training for professional female netball players.
